The aim of the research is to isolate, screen and study the phosphorus mobilizing activity of local strains of cotton rhizosphere bacteria. The qualitative test for acid formation of isolated rhizosphere bacteria was tested on Pikovskaya medium with tricalcium phosphate and indicator. Fifteen rhizobacterial isolates were isolated from cotton rhizosphere in the flowering-fruiting phase into pure culture. As a result of the qualitative test, out of 15 isolated cotton rhizosphere isolates, 6 isolates were the most active in acid formation in the early incubation period; their acid formation was 80-90% already on the 1st day of the experiment. 100% acid formation in these cultures was observed on the 3rd day of the experiment. As a result, by MALDI TOF mass spectrometry method, the species affiliation of active isolates was determined, No. 1С and 3С belong to Enterobacter cloacae, isolate No. 4С - to Bacillus subtilis, isolate No. 6С - to Pseudomonas aeruginosa, isolate No. 7С - to Pseudomonas chromraphis, and isolate No. 8С - to Bacillus atropaeus. In the next series of experiments, the total amount of titratable acids by the selected active acid formers was determined. The obtained results confirmed the data of qualitative acid formation test. The cultures of P. chlororaphis 7С and B. atrophaeus 8С were the most active acid generators relative to the other cultures, with total acidity ranging from 12.6±0.21 to 21.0±0.26 ml/100 mM NaOH. Thus, the 2 most active cultures of cotton rhizobacteria, P. chlororaphis 7С and B. atrophaeus 8С, were selected from 6 active cultures for further studies as a result of screening for acid-forming activity. |
